Biotium Human Lambda Light Chain (B-Cell Marker) (LLC/1738), CF488A conjugate, 0.1mg/mL
This MAb is specific to lambda light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the lambda light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. HHV 8 encodes a latent nuclear antigen (LNA), which is the product of the viral gene orf 73. LNA is capable of forming a complex with retinoblastoma susceptibility gene product, which may be related to its oncogenic activity. HHV8 is associated with three different diseases observed in AIDS patients; kaposi's sarcoma, primary effusion lymphoma (which is a rare type of non-Hodgkin lymphoma affecting the body cavities) and multicentric Castleman's disease. HHV 8 is the likely etiological agent of Kaposis sarcoma. Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Recognizes a protein of 30-33 kDa, which is identified as CD20 (Workshop V; Code CD20.4). It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. The protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B-cells and its cytoplasmic domains are differentially phosphorylated upon activation. This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk's and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Its expression is induced by the wild type, but not mutant, p53 suppressor protein. Normal cells generally display a rather intense nuclear p21 expression. Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb cocktail recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. Recognizes a protein of 94 kDa, which is identified as the glucose-regulated protein 94 (grp94) and also tumor rejection antigen (gp96). Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp s are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. This antibody reacts with Ig domain of CD47 protein. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. This antibody recognizes a 32 kDa protein, which is identified as homodimer of galectin-13 (also known as PP13). Galectins are a family of soluble beta-galactoside-binding lectins that modulate cell-to-cell adhesion and cell-to-extracellular matrix (ECM) interactions and play a role in tumor progression, pre-mRNA splicing and apoptosis. One such member, galectin-13, is a 139 amino acid protein that contains one galectin domain. Possessing lysophospholipase activity, galectin-13 exists as a disulfide-linked homodimer. Galectin-13 is suggested to have a developmental role in the placenta and may display immunobiological activity in feto-maternal blood-spaces. Biotium MUC5AC (Mucin 5AC / Gastric Mucin)(9-13M1), 1mg/mL
This MAb recognizes the peptide core of gastric mucin M1 (recently identified as Mucin 5AC). Its epitope is located in the N-terminal cysteine rich part of the peptide core of MUC5AC, which is heavily glycosylated. Its epitope is destroyed by beta-mercaptoethanol but not by periodate treatment. MAb 2-11M1 reacts with the protein backbone exclusively; it only reacts with fully deglycosylated MUC5AC. Therefore, the material under test should also be fully deglycosylated. This can be achieved with standard periodate oxidation method. The success of the deglycosylation can be checked with routine PAS (Periodic Acid Shiff) staining. After deglycosylation, the preparation should no longer be stainable with PAS reagent. Only then 2-11M1 will react should MUC5AC be present. This antibody reacts with a 70 kDa membrane-bound isozyme (Regan and Nagao type) of Placental Alkaline Phosphatase (PLAP) occurring in the placenta during the 3rd trimester of gestation. It is highly specific for PLAP and shows no cross-reaction with other isozymes of alkaline phosphatase. Anti-PLAP This antibody reacts with germ cell tumors and can discriminate between these and other neoplasms. Somatic neoplasms e.g. breast, gastrointestinal, prostatic, and urinary cancers may also immunoreact with antibodies to PLAP. Anti-PLAP positivity in conjunction with anti-keratin negativity favors seminoma over carcinoma. This MAb recognizes a protein of 134 kDa, which binds fibroblast growth factor and E-selectin (cell-adhesion lectin on endothelial cells mediating the binding of neutrophils). GITR (glucocorticoid-induced TNFR-related gene) is a member of the TNF-receptor superfamily, also known as TNFRSF18. It is expressed at low levels on resting T lymphocytes and at high levels on CD25 CD4 Tregs. The expression of GITR on T cells can be upregulated upon activation.
